Resumen
Life is not possible without the intervention of microorganisms and, given its importance, have been created to preserve biological collections and they can be used in different fields of science and technology. Exists variety of methods of preservation, in this work was determined the state of cryopreservation five bacterial strains by viability testing, standardizing and further implementing the system freeze-drying, taking as a starting point to them, evaluating them against three protective compounds. An evaluation of preservation methods on the basis of three indicators, viability, morphological stability and purity was performed; finding cryopreservation system implemented in the laboratory is optimal and glycerol 67% v/v works for almost all strains for long periods of time; sucrose 10% w/v lyoprotectant proved to be the best of the three evaluated, designating it as a general lyoprotectant for the laboratory. It was created a manual for conservation laboratory strains, including cryopreservation and freeze-drying protocols and safety of themselves. This work contributes to the field of microbiology conservation and is a stepping stone for registration Gene Bank Microbiology Laboratory of the Faculty of Environment and Natural Resources of the University District Francisco Jose de Caldas (CM-UDFJC) before the relevant national and international institutions.
