Resumen
Both fluorescence (F) and optical density (OD) are physical effects resulting from the light-matter interaction and have demonstrated their experimental importance in different fields of science. The devices that measure these effects have been used to evaluate chemical and biological variables; For example, fluorescence has become one of the few tools to estimate gene expression in living organisms. On the other hand, optical density is used to record the behavior of the bacterial population over time. The purpose of this internship was to carry out the characterization of a low cost team, created at the Universidad de Los Andes, describing a calibration methodology that includes, protocols and measurements with different substances such as Calcein for the calibration of F and McFarlan standard. for OD calibration. In addition, use concentrations of E-coli bacteria with fluorescent proteins under the action of a constitutive promoter and compare their measurements with standard commercial equipment.
