Ítem Contribución Al Estudio De Transportadores En Tripanosomátidos: Clonación De Por Lo Menos Un Gen Codificante Para Proteínas Transportadoras De Nad+Morales Herrera, David Santiago; Ayala Fajardo, Adis; Rubiano, Claudia ConsueloCutaneous Leishmaniasis is mainly caused by the parasite Leishmania braziliensis. The OMS estimates that occur between 0.7 and 1.3 million new cases per year therefore it has become a public health problem. For its study, this paper aimed to characterize in silico 3 candidates to NAD+ transporters in Leishmania braziliensis by primary structure analysis, secondary structure prediction and obtaining predictive models of tertiary structure, with which 19 residues conserved in the three candidates are identified, of them two residues are related to the transport of adenine nucleotides, also a high probability to present a regulation by phosphorylation/dephosphorylation but not by acetylation/deacetylation was found. Additionally, the presence of the domain of the family of "Mitochondrial Carriers" characterized by α helices transmembrane 6 was checked. By models from tertiary structure high identity was found with ADP/ATP mitochondrial transporter of the Bos taurus specie, suggesting a possible functional homology and therefore the ability to transport adenine nucleotides specifically NAD+ as ATP/ADP translocase of Chlamydia trachomatis. In addition, the presence of the genes of the three candidates with sizes of 1089, 1014 and 957 bp was checked in the genome of Leishmania braziliensis by experimental methods and recombinant proteins for each were obtained. These results provide strong evidence for further studies to verify their functionality.
