Diseño y síntesis de un péptido con y sin restricción conformacional de una proteína expresada por el Gen Pb-1 de Oryza sativa involucrada en el mecanismo de defensa contra la pyricularia

Resumen
The fungus Pyricularia Oryzae is the causative agent of the antagonist, disease of high impact in the production of rice (Oryza sativa) in the world, causing serious injuries on the ground and partial or total loss of the grain. The natural reaction of plants to pathogenic species is of short duration due to biotic and abiotic conditions of cultivation; however, this defense mechanism has promoted the study and development of resistant varieties, which are used for the control of the disease. With the vision to contribute to the expansion of the germplasm on resistance of rice varieties, the National Federation of Colombia rice and research groups Biomolc and UD proteome of the Universidad Distrital Francisco José de Caldas, have studied genes that confer resistance to the Pyricularia, as it is the case of the gene Pb - 1 of Oryza sativa, which expresses a protein involved in the mechanism of defense against this disease resistance. In this work were selected fragments LHEDQYAQILQE and DEEEKDNDDN with trend Alpha helical of this protein, is synthesized linear sequences and their corresponding analogues with conformational restriction of Alpha Helix modeled Dimer double (in patent process). The results by liquid chromatography high efficiency reverse phase (RP-HPLC), mass spectrometry (MS) and Dichroism circular (CD) show that the proposed products could be obtained and demonstrated the usefulness of the site of nucleation JAZ (in process of patent) to stabilize fragments of proteins with helical trend.
